In San Francisco, Jill Valentine is dealing with a zombie outbreak and a new T-Virus, Leon Kennedy is on the trail of a kidnapped DARPA scientist, and Claire Redfield is investigating a monstrous fish that is killing whales in the bay. Joined by Chris Redfield and Rebecca Chambers, they discover the trail of clues from their separate cases all converge on the same location, Alcatraz Island, where a new evil has taken residence and awaits their arrival.

Plot Summary:
In 1998, the Umbrella Corporation hired a private military firm to evacuate high-ranking Umbrella executives and government officials during the Raccoon City incident. Unit 6 was deployed, but most were bitten by zombies. Survivors JJ and Dylan Blake sought medical assistance but were instructed to quarantine and eliminate their fellow comrades. In 2015, Field Operations Support Agent Ingrid Hunnigan assigned DSO agent Leon S. Kennedy to apprehend DARPA researcher and robotics engineer Doctor Antonio Taylor in San Francisco for selling classified information to foreign powers. Leon tries to intercept them but is thwarted by Maria Gomez, a criminal who survived their last encounter the year prior.

Bioterrorism Security Assessment Alliance advisor Rebecca Chambers informs Agent Chris Redfield of twelve deaths from an advanced strain of the T-virus. Chris and Jill Valentine discuss their findings, which reveal an orca infected with the same strain swimming near the Greater Farallones National Marine Sanctuary. Jill and the Redfields travel to Alcatraz Island, where they discover blueprints for mosquito-like drones used to infect people with the virus. Dylan uses the drones to infect the Redfields and Leon, sparing Jill to witness her friends die. He later reveals he has taken over arms dealer Glenn Arias' crime syndicate and joined forces with Maria, who seeks revenge for her father Diego's death.

The Redfields, Jill, Leon, and Rebecca fight Dylan, who merges himself with a bio-organic weapon and overpowers the group. Claire and Rebecca take control of Dylan's drones and program them to infect him, weakening him enough for Jill, Leon, and Chris to kill him.
